The treatment of women with HER2-positive metastatic breast cancer has been reconfigured based on the results of a trilogy of randomised clinical trials published between 2012 and 2014.1–3 These studies led to the approval of the monoclonal antibody pertuzumab with trastuzumab and chemotherapy in patients with newly diagnosed metastatic disease, and of the antibody–drug conjugate trastuzumab emtansine in patients with trastuzumab-resistant metastatic disease.
