One of the first molecular targets for the treatment of non-small-cell lung cancer was the EGFR pathway. EGFR is overexpressed in non-small-cell lung cancer and other malignancies, whereas mutations in the intracellular domain of the receptor occur almost exclusively in lung cancer, especially adenocarcinomas. Irrespective of these differences, activation of the EGFR pathway results in a cascade of intracellular events leading to cell proliferation, metastases, angiogenesis, and reduced apoptosis.
